A novel hot-plate test sensitive to hyperalgesic stimuli and non-opioid analgesics BJMBR

It is widely accepted that the classical constant-temperature hot-plate test is insensitive to cyclooxygenase inhibitors. In the current study, we developed a variant of the hot-plate test procedure (modified hot-plate (MHP) test) to measure inflammatory nociception in freely moving rats and mice. Following left and right hind paw stimulation with a phlogogen and vehicle, respectively, the animals were placed individually on a hot-plate surface at 51ºC and the withdrawal latency for each paw was determined simultaneously in measurements performed at 15, 60, 180, and 360 min post-challenge. BACKGROUND: Algesia and inflammation are related with several pathological conditions. It is known that many drugs available for the treatment of these problems cause unwanted side effects. This study was aimed at evaluating acute toxicity and anti-inflammatory activity of Lampaya medicinalis Phil. (Verbenaceae) widely used in the folk medicine of Northern Chile against rheumatism, arthritis and body joints pain. RESULTS: Oral administration of hydroalcoholic extract (HAE) at the highest dose of 3000 mg/ Kg body weight resulted in no mortalities or evidence of significant behavioral changes. Analgesic and anti-arthritic effect of Corallocarpus epigaeus ABCL

Rheumatoid arthritis is a chronic inflammatory joint disease associated with the development of oxidative stress and inflammation. The safety and efficacy profile of 85% methanolic extract of Corallocarpus epigaeus (CE) was evaluated in the present study. In safety profile LD50 value was determined by carrying out an acute toxicity study. In efficacy profile, the analgesic activity was evaluated by both hot plate and tail immersion tests. The anti-inflammatory activity was assessed by carrageenan-induced paw edema and anti-arthritic effect by complete Freund's adjuvant induced arthritis. Effect of acupuncture on TNF-alpha, IL-1b and IL-10 concentrations in the peritoneal exudates of carrageenan-induced peritonitis in rats Ciência Rural

Acupuncture is an ancient and empirical therapeutic procedure known by its efficacy in the treatment of pain. However, the influence of acupuncture on inflammatory process is still poorly understood and additional research is needed. In this work, we investigated the mechanism of action of manual acupuncture on the inhibition of neutrophil migration to the peritoneal cavity induced by the inflammatory stimulus carrageenan in Wistar rats. Previous results from our laboratory showed that this anti-inflammatory effect is not due to endogenous corticoid release. About half of seaweed biomass is composed of polysaccharides. Most of these complex polymers have a marked polyanionic character. For instance, the red algal cell wall is mainly composed of sulfated galactans, agars and carrageenans, while brown algae contain alginate and fucose-containing sulfated polysaccharides (FCSP) as cell wall polysaccharides. Some marine heterotrophic bacteria have developed abilities to grow on such macroalgal polysaccharides. This is the case of Pseudoalteromonas carrageenovora 9(T) (ATCC 43555(T)), a marine gammaproteobacterium isolated in 1955 and which was an early model organism for studying carrageenan catabolism. Hind limb ischemic preconditioning induces an anti-inflammatory response by remote organs in rats BJMBR

Ischemic preconditioning (IPC), a strategy used to attenuate ischemia-reperfusion injury, consists of brief ischemic periods, each followed by reperfusion, prior to a sustained ischemic insult. The purpose of the present study was to evaluate the local and systemic anti-inflammatory effects of hind limb IPC in male Wistar rat (200-250 g) models of acute inflammation. IPC was induced with right hind limb ischemia for 10 min by placing an elastic rubber band tourniquet on the proximal part of the limb followed by 30 min of reperfusion. In vitro characterization and in vivo performance of mefenamic acid-sodium diethyldithiocarbamate based liposomes BJPS

Mefenamic acid (MFA) is a hydrophobic drug with low dissolution rate. This study aimed to develop stable and reproducible aqueous formulations of MFA using liposomes as drug carriers. The drug entrapment, particles size and drug release profiles, and stability and reproducibility of the liposomes were determined. In addition, the maximum tolerated dose (MTD) was determined in rats via the oral and intraperitoneal routes of administration. Also, the anti-inflammatory efficacy of these liposomes was evaluated using carrageenan-induced paw edema model in rats. Long-term ethanol intoxication reduces inflammatory responses in rats BJMBR

The anti-inflammatory effects of long-term ethanol intoxication were determined during ethanol treatment and withdrawal on the basis of neutrophil and eosinophil migration, hind paw edema and mast cell degranulation. Male Wistar rats (180-200 g, around 2 months of age) were exposed to increasing concentrations of ethanol vapor over a 10-day period. One group was evaluated immediately after exposure (treated group - intoxicated), and another was studied 7 h later (withdrawal group). Implantation of Walker 256 tumor decreases acute systemic inflammation in rats. Inflammatory hyperalgesia is one of the most important events of acute inflammation. The L-arginine/NO/cGMP/K+ATP pathway has been proposed as the mechanism of peripheral antinociception mediated by several drugs and physical exercise. The objective of this study was to investigate a possible involvement of the NO/cGMP/K+ATP pathway in antinociception induced in Walker 256 tumor-bearing male Wistar rats (180-220 g). Restorative Effects of A Newly Synthesized Peptide, Obiopeptide-1, in Cyclophosphamide- or Carrageenan-pretreated Mice Infected with Opportunistic Bacteria OAK

Large numbers of cyclophosphamide- or carrageenan-pretreated immunosuppressed adult ddY mice died within 10 days after inoculation with several species of opportunistic bacteria, including P. aeruginosa, K. pneumoniae, E. coli, S. aureus and methicillin resistant S. aureus. When immunosuppressed mice were administered bacteria in combination with two 100 μg/mouse intramuscular doses of a newly synthesized peptide, Obiopeptide-1 (OP-1), survival rates increased significantly.
